- Structure
- Monomer, belongs to the peptidase M10A family. 469 amino acids, predicted molecular weight of 54 kD.
- Distribution
- Extracellular. MMP-1 is expressed by fibroblasts, keratinocytes, endothelial cells, monocytes, and macrophages.
- Function
- MMP1 cleaves collagen of types I, II, III, VII and X. MMP-1 is thought to play an important role in the pathogenesis of rheumatoid and osteo-arthritis as mediators of cartilage and bone destruction. It is also involved in angiogenesis, tissue remodeling and cancer metastasis. MMP-1 regulates chemokine activity. MMP1 is inhibited by TIMPs, ?±2-macroglobulin (inhibitor of MMPs in bodily fluids). TNF-alpha; down-regulates TIMPs and enhances the expression of MMPs.
